Fig.1. Identification of protein interactions with the Matchmaker LexA Two-Hybrid System (Clontech). Plasmids containing the lacZ reporter gene (blue), the tested gene (green), and a library of putative interaction partners are propagated in yeast cells (EGY48) on selective media. Interaction of a fusion protein, which is bound to the lexA operator (pink) via a DNA-binding domain (black) with a second fusion protein possessing a transcription activating domain (purple) leads to detectable induction of the reporter gene.
