酪氨酸激酶HER2 和HER3与癌症的治疗
某些酪氨酸激酶在很多癌症中过度活跃，抑制它们的药物如慢性骨髓性白血病治疗药物imatinib可以成功使用，但它们对所有由酪氨酸激酶驱动的癌症并不都起作用，新的研究工作指出了可能的原因。一种特定的激酶，即HER2，在乳腺癌中经常过度活跃，它通过该激酶家族中的另一个成员HER3传递信号。Sergina等人发现，当HER2被激酶抑制药物抑制时，一种反馈机制就会造成胞质膜中活性HER3的增加，如果HER2活性没有被彻底阻断的话，HER3将在胞质膜中继续发出癌细胞增殖的信号。所以，能够彻底阻断HER2活性（也降低HER3活性）的更有效的抑制药物也许能更有效地治疗癌症。
