Does anybody know how to isolate single-stranded DNA from mammalian cells?

I don't know if you have obtained an answer to your question yet, but single stranded DNA can be isolated from mammalian cells with Chelex 100, a chelating resin that has a high affinity for polyvalent metal ions. Used during the boiling of DNA, it prevents the breakdown of DNA at high temperatures in low ionic strength solutions. It can be purchased from Sigma (800-325-3010).
