1.Excise band of interest from a TAE gel; estimate volume by weighing the gel slice.
2.Dissolve the gel slice in 3 volumes of NaI (from Geneclean kit)at 55E C for 5 min or until the gel dissolves.
3.Add 1 ml of Wizard miniprep resin,10 min.at room temperature.
4.Add DNA resin mixture to a syringe and pass through a minicolumn.
5.Wash with 2 ml of Wizard wash buffer; spin 2 min in a microfuge.
6.Elute with 70EC TE or water (50 μl),2 min in a microfuge.
